A quadrupole ion trap mass spectrometer MSn, with ESI and APCI sources, coupled with a Liquid Chromatography system, is an equipment of the Center for Pharmaceutical Studies (CEF), a university research unit, and is installed in the Laboratory of Mass Spectrometry (LEM/UC).

This service (LEM/UC) closely supports the interdisciplinary research in Health Sciences developed in the Center for Pharmaceutical Studies and the research in other centers of the University of Coimbra. Specifically, LEM/UC gives technical and scientific support to the discovery and development of natural and synthetic bioactive compounds, analysis of xenobiotics, drug metabolism studies and analysis of cosmetics and dosage forms.

Beyond the research goals, LEM/UC gives training to researchers in this area and is also open for external services.

Furthermore, LEM/UC is integrated in a national mass spectrometry network (RNEM), sharing its scientific and technical capabilities within the nine units involved.

The equipment has been financed by FCT under the scope of the “Programa Nacional de Reequipamento Científico”.

Equipment

Quadrupole/Ion Trap Mass Spectrometer (QIT-MS)

coupled to a Liquid Chromatograph of High Performance THERMO FINNINGAN

Financed by Fundação para a Ciência e Tecnologia, under the scope of the Programa Nacional de Reequipamento Científico

This equipment is integrated in the National Mass Spectrometry Network, Rede Nacional de Espectrometria de Massa (RNEM).

Mass Spectrometer Detector

LC/MS analyzer ultra-sensible “QUADRUPOLE/ION TRAP” with multiple MS/MS (MSn) and for masses from 50 to 2000 (expandable in a virtual way to 8000 a.m.u. for ESI).

- Ion sources available
- Atmospheric Pressure
- Ionization modes
- ESI (electrospray ionization)
- APCI (atmospheric pressure chemical ionization)

The detectors provide five levels of analysis:

Mass analysis (molecular weight information)
- Two-stage mass analysis (structural information)
- Wideband activation (structural information)
- Zoom Scan analysis (high resolution scan)
- Compound analysis after LC.

Liquid Chromatography System

coupled to the Mass Spectrometer for MS/MS and MSn

  • LCQ Advantage
  • Ion Max
  • MSMS

LCQ Advantage is a system based on Quadrupole/Ion Trap technology, allowing qualitative and quantitative analyses in complex matrices, with mass accuracy (< 0.1 a.m.u.) and high sensitivity.

- Diode Array Detector (190-800nm) a 5 cm cell <> increase of sensitivity
- Autosampler with preparation and dilution of the samples
- MSPUMP for micro-flows.

Applications

  • Structural elucidation of organic and bio-organic compounds either pure or in complex matrices
  • Control/optimization of chemical processes and purification methods
  • Pharmaceutical and cosmetic analysis
  • Metabolism studies
  • Different complex matrices can be analysed, namely plant extracts, foodstuff, biological fluids, pharmaceuticals, waste waters, river waters.
